San Diego FC was formed almost two years ago and now consists of a total of 30 clubs in MLS.
To get San Diego into the league has been a long-term goal, said MLS chief Don Garber before the match.
The team's big star is Hirving Lozano, who has played for Mexico in the two most recent World Championships. He signed with San Diego last summer, but belonged to Dutch PSV Eindhoven until the turn of the year.
San Diego is the tenth club to join MLS since 2017.
