Mitoma, who injured one of his thigh muscles in connection with the Premier League match between Brighton and Wolverhampton, is not included in the World Cup squad that has now been presented.
Mitoma scored Japan's goal in the victory against England in March.
Another player missing is Takumi Minamino, who plays for Monaco. He has been out of action since December last year with a knee injury. Japan opens the World Cup against the Netherlands on June 14 and faces Sweden on June 26 in Dallas, Texas.
